ABSTRACT

Objective To observe the effect of miR-369-3p on vascular endothelial growth factor C (VEGFC) gene in bladder cancer cells EJ and 5637 and observe its effect on cell proliferation and apoptosis. Methods Bladder cancer cell lines EJ and 5637 were transfected with miR-NC (control group) or transfected with miR-369-3p (experimental group). The target gene of miR-369-3p was predicted by Targets can target gene prediction software.Fluorescence real-time quantitative polymerase chain reaction (qPCR) was used to detect the changes of VEGFC at mRNA level. The changes of VEGFC,p-Raf-1 and phosphorylated extracellular signal-regulated kinase 1/2 (p-ERK1/2) at the protein level were detected by Western blotting. The effect of miR-369-3p on the proliferation of bladder cancer cells was detected by cell counting kit (CCK-8) and clone formation experiment. The effect of miR-369-3p on apoptosis was detected by flow cytometry. Results After transfection with miR-369-3p,the expression of VEGFC at mRNA and protein levels was significantly decreased (P < 0.05),the expression of p-Raf-1 and p-ERK1/2 protein was significantly decreased, the cell proliferation ability was significantly decreased (P < 0.05),the number of clones formed was significantly decreased (P < 0.05) and the apoptosis was significantly increased (P < 0.01).Conclusions miR-369-3p can inhibit the proliferation and promote the apoptosis of bladder cancer cells by interfering with the expression of VEGFC gene, which may provide a new target for the biological therapy of bladder cancer.

ABSTRACT

OBJECTIVETo investigate the influences of temperature, lightness, storage method, storage time, and gibberellin on seed germination of Gentiana rigescens.

METHODThe germination rates of G. rigescens in different treatments were observed.

RESULT AND CONCLUSIONThe most suitable temperature for the seed germination was 25 degrees C, at which the germination rate was 76.33%. The effect of lightness on the seeds was significantly; the germination rate of the seed was very low. Under the natural condition, the best storage method was dry storage (within 6 months), which could promote the after-ripening of the seed. 100-1 000 mg x L(-1) gibberellic acid could significantly reduce the seed germination time, and 500 mg x L(-1) gibberellic acid increased the germination rate of the seed to 95.00%.

ABSTRACT

OBJECTIVEThe interrelation of yield and agronomic traits of Gentiana rigescens was studied for the germplasm and breeding variety of this species.

METHODTwelve agronomic traits, root diameter, root length, root number, root biomass, stem diameter, plant height, the first branch number, leaf length, leaf width, leaf length/leaf width ratio, calyx length, and calyx number of G. rigescens from 26 wild populations in Yunnan were determined for correlation analysis, multiple stepwise regression analysis and path analysis.

RESULTCorrelation analysis showed that there were significantly positive correlation between the traits of aboveground part and the length, diameter, number, and biomass of the root. Multiple stepwise regression analysis showed that length, width, and number of root, plant height, the first branch number, and the calyx number were the main factors that affected the root biomass. Path analysis showed that the diameter, length, and number of the root, the stem height, and the first branch number had a direct positive effect on the root biomass.

CONCLUSIONThe traits, such as high and strong stem, high number of first branch number and shrubby shape could be selected for the breeding and high yielding of G. rigescens.

ABSTRACT

To investigate the effect of hSCGF-alpha on human Umbilical Cord Mesenchymal Stem Cells (hUCMSCs), we obtained hSCGF-alpha using genetic engineering, hSCGF-alpha gene was amplified from hUCMSCs cDNA using two-step PCR and was inserted into pET-28a(+) plasmid vector. Induced by IPTG at 20 degrees Celsius for 24 h, the fusion protein expressed in E. coli BL21 (DE3) was mainly existing in soluble form. The recombinant hSCGF-a was purified using NI-NTA affinity chromatography and the purity was up to 90%. The colony forming test revealed that combined use hSCGF-alpha and rmGM-CSF (recombinant murine GM-colony stimulating factor, rmGM-CSF) had granulocyte/macrophage (GM) promoting effects on murine bone marrow GM progenitor. In addition, the results indicated that hSCGF-alpha and rhGM-CSF had stimulatory effect on hUCMSCs and their synergetic effect was the strongest.

ABSTRACT

OBJECTIVE:To investigate the methods of calculating pediatric dose of medication.METHODS:The validity of several methods of calculating pediatric dose of medication was evaluated by statistical methods and demonstration analysis, and the expression of the pediatric dosage of administration in drug instruction was discussed.RESULTS&CONCLUSION:There exist improper medical manipulation in the rational use of medicines in pediatrics, uniform criterion and calculation are lack for measuring age groups in pediatrics, aseries of integrated and uniform criteria to determine the safe dosage for child are needed.

ABSTRACT

OBJECTIVE:A pharmacoeconomical analysis was performed to evaluate three therapeutic schemes for preventing post-appendectomy infections in children METHODS:The study included 119 children undergoing appendectomy According to the pathological results of appendix, they were divided into group Ⅰ and group Ⅱ,and treated with three different schemes respectively The schemes consisted of A:cefazolin+benzylpenicillin+metronidazole,B:cefotaxime+benzylpenicillin+metronidazole and C:cefazolin+benzylpenicillin+metronidazole+claforan These three schemes were assessed with pharmacoeconomic evaluation RESULTS:As for group Ⅰ,scheme A and B were better than scheme C As for group Ⅱ,scheme A was the best CONCLUSION:Application of pharmacoeconomics will optimize therapeutic scheme and guide rational use of drugs
